
BASIC linkers are used to physically connect bioparts in the BASIC assemby method.
5 different classes of linkers are available:

Neutral Linkers connect DNA parts with neutral sequence (45bp).

Methylated Linkers encode BASIC prefix and suffix in methylated form, so that multiple bioparts flanked by these linkers constitute a new single BASIC biopart after the assembly.

RBS linkers contain 1 of 3 defined 5'-UTR regions combined with 1 of 15 RBS sequences.

Mixed RBS Linkers encode for RBS pools of 12, 24 or 36 specific RBS sequences.

Peptide Fusion Linkers connect flanking bioparts in frame creating fusion proteins.
Each BASIC linker consists of 4 oligos, all of which are HPLC purified for highest assembly fidelity. Then 2 oligos are pre-annealed to constitute linker suffix and prefix section respectively.
